Brain Ciliary Neurotrophic Factor (CNTF) and hypothalamic control of energy homeostasis

چکیده

Obesity is a chronic, metabolic disease of complex and multiple causes leading to an imbalance between energy intake and output, and to the accumulation of large amounts of body fat. It is caused by inherited as well as acquired factors, including excessive food intake, sedentary lifestyle and unhealthy eating habits. During the past 20 years, obesity among adults has risen significantly with urbanization, economic development and market globalization. According to the World HealthOrganization (WHO) statements, more than one billion people worldwide are overweight or suffer from obesity, and the number of affected children has more than doubled since 1980 in the USA and Europe. In France, the latest data from Roche show that overweighting and obesity affect, respectively, more than 30% and 14.5% of adults (ObEpi-Roche, 2009). But far more worrying are the increasing and acceleration of this problem into developing countries and, based on current trends, it is predicted that the levels of obesity will continue to rise unless action is taken now (McLellan, 2002). The consequences of obesity for adults are well known. Obesity contributes to the development of many diseases, including diabetes, hypertension, dyslipidemia (for instance, high total cholesterol or high levels of triglycerides), stroke, cardiovascular disease, and some cancers (Abelson and Kennedy, 2004). As a result, the obesity epidemic has prompted important efforts to develop safe and potent therapies. However, currently approved drugs for obesity such as appetite suppressants have limited efficacy and act acutely, with patients rapidly regaining weight after the cessation of treatment. The neurocytokine ciliary neurotrophic factor (CNTF) seems to deviate from this paradigm since its administration to rodents or patients maintains lowered body weights several weeks after terminating treatment (Lambert et al., 2001).

The Cytokine Ciliary Neurotrophic Factor (CNTF) Activates Hypothalamic Urocortin-Expressing Neurons Both In Vitro and In Vivo

Ciliary neurotrophic factor (CNTF) induces neurogenesis, reduces feeding, and induces weight loss. However, the central mechanisms by which CNTF acts are vague. We employed the mHypoE-20/2 line that endogenously expresses the CNTF receptor to examine the direct effects of CNTF on mRNA levels of urocortin-1, urocortin-2, agouti-related peptide, brain-derived neurotrophic factor, and neurotensin....

gp130 signaling in proopiomelanocortin neurons mediates the acute anorectic response to centrally applied ciliary neurotrophic factor.

Ciliary neurotrophic factor (CNTF) exerts anorectic effects by overcoming leptin resistance via activation of hypothalamic neurons. However, the exact site of CNTF action in the hypothalamus has not yet been identified. Using Cre-loxP-mediated recombination in vivo, we have selectively ablated the common cytokine signaling chain gp130, which is required for functional CNTF signaling, in proopio...

Ciliary neurotrophic factor corrects obesity and diabetes associated with leptin deficiency and resistance.

Receptor subunits for the neurocytokine ciliary neurotrophic factor (CNTF) share sequence similarity with the receptor for leptin, an adipocyte-derived cytokine involved in body weight homeostasis. We report here that CNTF and leptin activate a similar pattern of STAT factors in neuronal cells, and that mRNAs for CNTF receptor subunits, similarly to the mRNA of leptin receptor, are localized in...

The anorexigenic cytokine ciliary neurotrophic factor stimulates POMC gene expression via receptors localized in the nucleus of arcuate neurons.

Ciliary neurotrophic factor (CNTF) is a neural cytokine that reduces appetite and body weight when administrated to rodents or humans. We have demonstrated recently that the level of CNTF in the arcuate nucleus (ARC), a key hypothalamic region involved in food intake regulation, is positively correlated with protection against diet-induced obesity. However, the comprehension of the physiologica...

Opposite effects of a high-fat diet and calorie restriction on ciliary neurotrophic factor signaling in the mouse hypothalamus

In the mouse hypothalamus, ciliary neurotrophic factor (CNTF) is mainly expressed by ependymal cells and tanycytes of the ependymal layer covering the third ventricle. Since exogenously administered CNTF causes reduced food intake and weight loss, we tested whether endogenous CNTF might be involved in energy balance regulation. We thus evaluated CNTF production and responsiveness in the hypotha...

Neurogenesis in the hypothalamus of adult mice: potential role in energy balance.

Ciliary neurotrophic factor (CNTF) induces weight loss in obese rodents and humans, and for reasons that are not understood, its effects persist after the cessation of treatment. Here we demonstrate that centrally administered CNTF induces cell proliferation in feeding centers of the murine hypothalamus. Many of the newborn cells express neuronal markers and show functional phenotypes relevant ...
